On May 19, 2012, Killorn concluded his playing career with the Harvard Crimson men’s ice hockey team and signed a two-year, entry-level contract with the Tampa Bay Lightning.

He earned his first NHL assist on February 10, 2013, against the New York Rangers, on a goal by Vincent Lecavalier. On February 17, 2013, against the Florida Panthers, a week later, Killorn scored his debut goal. With 48 games remaining in the lockout-shortened season, Killorn concluded his first year with 7 goals and 12 assists in 38 contests.
In a 4-2 loss to the St. Louis Blues on March 4, 2014, Killorn participated in his 100th NHL game and scored a goal in the opening frame.
Killorn played in all 82 games of the 2013–14 season, his second in the NHL, and finished with 17 goals, 24 assists, and 41 points.
On April 16, 2014, Killorn made his playoff debut in a 5-4 loss to the Montreal Canadiens. He scored a goal against Carey Price and assisted on a goal by team captain Steven Stamkos. Killorn was re-signed by the Lightning to a two-year contract deal on June 19, 2014.
In a 5-1 victory over the Arizona Coyotes on October 28, 2021, Killorn registered his 200th NHL assist on a goal by Andrej Sustr. Killorn played his 700th NHL game on March 13, 2022, in a 2-1 victory over the Vancouver Canucks.
Killorn scored his 400th NHL point on April 21, 2022, as the Toronto Maple Leafs were defeated 8-1. By participating in all 82 games once more and scoring 25 goals, 34 assists, and 59 points, Killorn maintained his durability. Killorn and the Lightning advanced to a second Stanley Cup Final, where they fell to the Colorado Avalanche in six games.
